EFFECTS OF CHIGUO OIL STEROL ON PROLIFERATION AND DIFFERENTIATION OF RAT PREADIPOCYTES

Abstract

To investigate effects of Chiguo oil sterol on proliferation and differentiation of rat adipocytes and to elucidate its potential mechanism,rat's primary preadipocytes invitro were cultured.Treated preadipocytes with 0μmol/L(control group),2 μmol/L,4 μmol/L,8 μmol/L Chiguo oil sterol.Cell proliferation was analyzed by MTT assay.The degree of adipogenesis and differentiation were measured by Oil Red O staining extraction assay and the expression of peroxisome proliferation activated receptor-γ2(PPARγ2)mRNA were detected by reverse transcriptase-polymerase chain reaction(RT-PCR).It was demonstrated that the optical density(OD)of MTT were decreased,especially treated by 4 μmol/L and 8 μmol/L.The OD of Oil Red O staining and the expression of PPARγ2 mRNA were all decreased after treated by 4 μmol/L and 8 μmol/L Chiguo oil sterol(P0.05).
