Abstract

[Objective]To explore the effects of genistein on proliferation of human glioma cell line U251MG and possible mechanism involved.[Methods]The effects of genistein on proliferation of U251MG cells were shown by MTT assay.The expression of p53(mutant),c-erbB1,c-myc,cyclinD1 proteins were examined with flow cytometry and immunocytochemistry.[Results]When genistein reached a certain concentration and/or a certain time,the growth of U251MG cells was inhibited in a dose-and time-dependant manner.When U251MG cells were treated with genistein for 48h,p53(mutant),c-myc proteins expression were significantly decreased compared with control group in a dose-dependent manner(F = 25.766,P﹤0.01;F = 22.974,P﹤0.01),while the c-erbB1,cyclinD1 had no obvious changes in protein expression(F = 0.984,P﹥0.05;F = 2.416,P﹥0.05).[Conclusion]Genistein can inhibit the proliferation of U251MG cells significantly,the antitumor mechanism may be related to the decreased protein expression of p53(mutant)and c-myc.
