Abstract

Objective To discuss the application in the treatment and prognosis of breast cancer with immunohistochemistry(IHC) and real-time quantitative PCR(RT-PCR) for breast cancer susceptibility gene Her-2、EGFR、CK5/6.Methods The Her-2,EGFR,CK5/6 gene expression levels were detected by IHC and RT-PCR in 41 patients with breast cancer.The sensitivity and specificity of two kinds of methods were analyzed and compared.Results Forty-one cases of breast cancer were women.Immunohistochemical methods showed 21 cases of Her-2 positive expression,8 cases of EGFR positive,and 8 cases of CK5/6-positive;But RT-PCR method showed Her-2 positive in 27 cases,EGFR positive in 21 cases,and CK5/6 positive in 21 cases.Her-2 positive rates were not statistically different with both of these methods(P0.05);EGFR,CK5/6 positive rates of different methods were statistically significant(P0.05).Conclusion Detection of EGFR and CK5/6 have difference in amplification and protein expression in different tissues,but Her-2 has not with two methods.RT-PCR method has higher sensitivity and accuracy,but their results are consistent.
