Abstract

The conditions for transformation of Escheriehia coli by isolated tetraeycline resistance plasmid pSC101 DNA are reported.Drug resistance is expressed in transformed cells.There are certain conditions which affect transformation.After cells take up DNA,a period of time is needed to express tetracycline resistance characterization.The optimal concentration of cells for transformation is 3.05×106 cells/ml.Cells will be transformed by plasmid DNA after treated with CaCl2 the optimal concentration of CaCl2 for transformation is 40 mM.As expected,the higher the concentration of plasmid DNA,the higher will be the frequency of transformation.The other factor for transformation is cell itself.After cells are grown to an optical density of 0,8-1.0 at 590nm,they possess the best conditions for transformation.
