Effects of Spironolactone on Ventricular Remodeling in Rats after Acute Myocardial Infarction

Abstract

Objective To explore the effects of Spironolactone on LV remodeling in post-MI rats. Methods Female rats having sham operation or left coronary artery ligation were killed 24 hours,1 week,2 weeks and 4 weeks after the operation,respectively. Oral administration of Spironolactone (40 mg/d) was given to the rats in drug group by gavage once a day from 3 hours to 4 weeks. Tap water was given to the rats in MI and sham-operation groups under the same experimental conditions. The changes of the left ventricular absolute weight (LVAW),left ventricular relative weight(LVRW),left ventricular transverse diameter and sphericity indices were analyzed. Results The infarction sizes were 23%-35%. After 1 week of the operation,the LVAW,LVRW and left ventricular transverse diameter increased markedly in MI group,compared with sham-operation rats,and the left ventricular remodeling happened 1 week after MI in rats. The above-mentioned indexes were improved markedly after 4 weeks of treatment with Spironolactone(P 0.05). Conclusion Using Spironolactone early after MI has protective effects on the left ventricular structure.
