Therapeutic Effects of Dihydrolycorine on Focal Cerebral Ischemia-reperfusion Injury in Rats

Abstract

Objective To investigate the therapeutic effects of dihydrolycorine(DL) on focal cerebral ischemia-reperfusion injury in rats.Methods The right middle cerebral artery ischemia models were established by ligating the internal carotid artery with a nylon suture for 1 h,following reperfusion for 24 h.SD rats were randomly divided into sham group,model group and different dosage of DL groups.DL(10,20,40 mg·kg-1) were respectively given at the beginning of cerebral ischemia immediately,11 h and 23 h after reperfusion through intraperitoneal injection.Normal saline were given in sham group and model group.The therapeutic effects of DL at different dosages were evaluated with neurological score,infract size,cerebral edema extent,myeloperoxidase(MPO) activity.Results The infarct size and cerebral edema extent were significantly decreased by giving DL after ischemia,the neurological function was significantly improved and the activity of MPO was significantly decreased correspondingly(P 0.05).Conclusion DL showed obvious therapeutic effects on cerebral ischemia reperfusion injury in rats and the effects may be related to the anti-inflammatory activity of DL.
