Abstract
Objective:To investigate the mechanism of anti-fibrosis action of Fuergan No.6(FEG6).Methods:Rats model was induced by in- traperitoneal injection of dimethylnitrosamine(DMN).The model rats in the treated group were treated with FEG6 once daily for 4 weeks to observe the liver pathology(with HE stain),collagen(with ponceau stain),superoxide dismutase(SOD)activity,malondialde- hyde(MDA)and hydroxyproline(Hyp)content in liver,serum level of alanine transaminase(ALT)and liver function,and compared with those in model rats treated with γ-interferon as positive control.The correlation analysis among Hyp,MDA and SOD was conduct- ed.Results:Collagen in liver tissue was markedly increased in the model rats with fibrous septa formation;serum ALT level raised,SOD activity in liver lowered significantly,and liver content of MDA and Hyp increased significantly.The MDA content was positively corre- lated with Hyp content,and SOD activity was negatively correlated with Hyp content.Both FEG6 and γ-interferon could reduce the col- lagen deposition in liver tissue significantly,lower serum ALT level,raise SOD activity and decrease the content of MDA and Hyp,no significant difference was found between the effects of the two treatments.Conclusions:DMN could arise lipid peroxidation in liver,and the DMN induced liver fibrosis is closely related with lipid peroxidation.FEG6 has good anti-lipid peroxidation effect,which is the im- portant mechanism of it in treating liver fibrosis.
