Abstract
In this study, ultrasonic assisted extraction method was used for extracting polysaccharide of Portulaca oleracea L.(POP),and Sephadex G-100 column chromatography was used for purification of POP.The optimal extraction conditions were obtained as follows: liquid-solid ratio 45∶1,ultrasonic power 130W,ultrasonic time 55min and ultrasonic temperature 75℃.Under these conditions, the average extraction rate of POP was 13.28%.Through Sephadex G-100 column chromatography, three sub-fractions were obtained. After the first sub-fractions were collected,it was found that the molecular weight distribution of POP-a was homogeneous, which is a kind of protein-containing acidic polysaccharides and not contain starch and reducing sugar.
