Abstract

Objective To observe the expression of reporter gene green fluorescent protein(GFP) in nasopharyngeal carcinoma(NPC) cell line CNE-2Z.Methods Control plasmid pPAT-GFP carried with GFP reporter gene and three different eukaryotic expression plasmids carried with GFP and aim gene teloRZ(pGFPuro-teloRZ 2.1?pGFPuro-teloRZ 7.1 and pGFPuro-teloRZ 7.7) were transfected into a NPC cell line CNE-2Z by electroporation.Then the expression of GFP in the live and the fixed transfected cells was tested by fluorescent microscopen and flow cytometry.Results Green fluorescence could be detected in intact viable cells but not in fixed cells.The rate of expression was affected by the insertion of the aim gene.Conclusion The GFP gene could be stably expressed in NPC cell line CNE-2Z transfected by plasmids containing aim gene.The GFP gene could be used as a reporter gene for plasmid transfection and gene expression in the research of CNE-2Z cells and ribozyme targeting the template region of telomerase RNA.
