Abstract
Eight isolating media including five basal media (LB medium,TSA medium,King's medium B,Starch ammonium medium,Czapek Dox Agar Modified medium) and three oligotrophic media (0.1×LB medium,YG medium,R2A medium ) were utilized to isolate endophytic bacteria from Phyllostachys edulis,aiming at screening the suitable media. In eight isolating media,the more numbers of bacteria were isolated from YG medium,King's medium B and R2A medium,1.31×106,8.03×105 and 6.45×105 CFU/gfw each other; while eight kinds of bacteria were isolated from R2A medium,seven kinds of bacteria were isolated from YG medium and King's medium B. The preponderant bacteria from any medium was isolated and purified,and the 16S rDNA samples were sequenced. The preponderant bacteria from LB medium,TSA medium,King's medium B,Starch ammonium medium,Czapek Dox Agar Modified medium,YG medium and R2A medium were closely related to Mycoplana ramose (91%); while the preponderant bacteria from 0.1×LB medium were closely related to Leifsonia poae (99%). The preponderant endophytic bacteria were Leifsonia poae from 0.1×LB medium and were Mycoplana sp. from the others. The results revealed that the isolation effect of R2A medium,YG medium and 0.1×LB medium there were better and they were the suitable media for isolating endophytic bacteria from Phyllostachys edulis.
