Influnce of losartan on pharmacokinetics of cyclosporin A in rabbit

Abstract

Objective To study the influnce of losartan on pharmacokinetics of cyclosporin A(CsA) in rabbit.Methods By fluorescence polarization immunoassay(FPIA method),determined blood concentration of CsA of six rabbits before and after combined losartan,and analysed pharmacokinetic parameters of two groups(single combination with CsA group and losartan group).Results The peak concentration(Cmax) of CsA of losartan group significantly increased,area under the curve(AUC0→24) increased significantly(P0.01),plasma clearance(CL) and decreased and apparent volume of distribution(V) significantly decreased(P0.05 or P0.01),the remaining pharmacokinetic parameters had no significant change(P0.05).Conclusion Combineing losartan can increase CsA blood concentration,it combineing CsA must monitor CsA concentration in blood,and ensure safe and effective treatment.
