Abstract
Objective To explore the feasibility of isolating and cultivating rat nucleus pulposus cells(NPCs)by the sequential enzyme method in vitro,providing a theoretical basis for obtaining high purity NPCs.Method Isolateing and cultivateing rat nucleus pulposus cells by the sequential enzyme method;observing cell adhesion,proliferation and cell morphology changes by inverted phase contrast microscope.Result Nucleus pulposus cells were isolated by the sequential enzyme method after repeated change medium and passage,and cells presented typical nucleus pulposus cells morphology,and were closely packed like whirlpool,island,colony.Conclusion The sequential enzyme method was able to isolate NPCs in higher purity,and it was an effective method culturing rat nucleus pulposus cells in vitro.
