Abstract

Objective To investigate how to establish a rat model for non-insulin-dependent diabetes mellitus(DM) rat model induced by streptozotocin(STZ) in combination with high-sugar and high-fat diets.Methods 40 healthy male SD rats were randomly divided into the high-fat diet group,the DM control group and the normal group.STZ(60mg/kg) was injected intraperitoneally into rats in the high-fat diet group and the DM control group.Rats in the high-fat diet group were fed with high-sugar and high-fat diets for 4 weeks,but rats in the DM control group were fed with normal diet for 4 weeks.Rats in the normal group were injected intraperitoneally with the same dose of citric acid buffer,and were fed with normal diet for 4 weeks.The fasting blood glucose,body weight and urine sugar were detected 72 hours,1,4,8 and 12 weeks later.Results Fasting blood glucose,body weight of rats in high-fat diet group or DM control group increased significantly than those in normal group(P0.05).High blood glucose in high-fat diet group could maintain longer than that in DM control group,and urine sugar in high-fat diet group was more positive than that in DM control group.Conclusion The rat model of DM could be successfully induced by STZ in combination with 4-week high-sugar and high-fat diets.
