Abstract

Objective To investigate the expression and significance of lipoprotein lipase(LPL) in hepatocyte steatosis model interfered in adiponectin.Methods A model of oleic acid-induced steatosis of L-02 strain hepatocytes(normal adult human hepatocytes) was established,and interfered in adiponectin.The accumulation of lipid droplets in hepatocytes was observed by light microscopy after oil red-0 staining.The mRNA and protein expression of lipoprotein lipase in hepatocytes were detected by reverse transcription and polymerase chain reaction(RT-PCR)and Western blot.Results More orange-red lipid droplets were found in the cytoplasm 72h in the intervention group(group T) after culture than steatosis group(group F).The expression of liproprotein lipase mRNA and protein was upregulated in group T,and the peak was reached in 24h.Compared with group F,the mRNA and protein expression of liproprotein lipase in Group F were significantly increased in the Group T(P0.01).Conclusion Adiponectin may promote the expression of liproprotein lipase in steatotic hepatocytes which may relieve hepatocyte adipose degeneration though decrease the synthesis of triglyceride.
