Abstract
Objective To investigate the inhibitory effect of Oridonin on leukemic NB4 cells and its mechanism. Methods Variais concentrations of Oridonin Were given to NB4 cells in vitro. The inhibitory ratio of the cells were measured by MTT assay,its apoptosis was observed by flow cytometry (FCM) and Hoechst 33258 fluorescence staining,and the activity of telomerase was detected by TRAP-PCR-ELISA before and after apoptosis. Results Oridonin could decrease the activity of telomerase,induce apoptosis and inhibit the growth of NB4 cells significantly .The effect was in a time-and dose-dependent manner. Conclusion Oridonin can inhibit the growth of NB4 cells and induce apoptosis and decrease of the telomerase activity of NB4 cells may be one of the most important mechanisms.
