Abstract
Object To investigate the inhibitory effect of oridonin on Raji cells (B lymphocyte leukemic strain) and its mechanism. Methods Raji cells in culture medium in vitro were given different concentrations of oridonin. The inhibitory ratio of the cells was measured by MTT assay, apoptosis of cells was observed by flow cytometry (FCM) and Hoechst 33258 fluorescence staining, and the activity of telomerase was detected using TRAP-PCR-ELISA before and after apoptosis occurred. Results Oridonin could decrease the activity of telomerase, as well as cause apoptosis and inhibit the growth of Raji cells significantly. The suppression was both in time- and dose-dependent manner. Conclusion Oridonin has anti-leukemic efficacy on B lymphocyte leukemia in vitro very significantly, inducing apoptosis and decreasing the activity of telomerase may be most important mechanism.
