Role of TNF-α、IL-8 in the lung injury induced by mechanical ventilation

Abstract

Objective Investigate the possible roles of tumor necrosis factor alpha(TNF-α),interlukin-8(IL-8)in the lung injury induced by mechanical ventilation.Methods Twenty-four normal healthy small pigs were given continuous different tidal volume as animal model,they were random divided into control group,low tidal volume group(group A),normal tidal volume group(group B),and high tidal volume group (group C), contents of TNF-α、IL-8 and myeloperoxidease (MPO) in serum and lung were observed with radioimmumoassay and enzyme linked immunosorbent assay and MPO measurent 1 d,3 d and 7 d following ventilation respectively.Results The contents of TNF-α、IL-8 and MPO in serum and lung were higher in group A、B and C than those in control group( P 0.05 or P 0.01).In addition,TNF-a value was at it's maximum 3 d later,IL-8 value and MPO value were at their maximum 7 d later,above change were significantly in group A and group C.Conclusion TNF-α、IL-8 may play important roles in the lung injury induced by mechanical ventilation. [
