Abstract

To obtain hybridoma cell lines secreting McAbs against SARS-associated coronavirus(SARS-CoV) S protein and N protein,Balb/c mice were immunized with the recombinant SARS-CoV S and N fusion protein.Spleen cells from Balb/c mice were fused with Sp2/0 myeloma cells.The specific McAbs were screened by ELISA and identified by western blot.Two hybridoma cell lines(1F11 and 3D2) secreting specific McAbs to SARS-CoV were selected and determined to recognize different epitops on S and N protein fusion protein.The titers of the two clones were as the following: 1:1024,1:512(supernant);1×10~(-6),2×10~(-5)(AF).McAbs 1F11 recognize the epitope on N protein,whereas 3D2 recognize the epitope on S protein.This result suggested that specific McAbs could be used for developing diagnosis reagent.
