The Impact of Vincristine on Cell Proliferation and Cell Cycle in Nasopharyngeal Carcinoma Cells

To investigate the impact of vincristine(VCR)on the cell proliferation and cycle in nasopharyngeal carcinoma cell line CNE-2Z.The inhibited rates of the proliferation and IC 50 values were determined by MTT assay.Cell cycle was analyzed by flow cytometry.Cells were treated with VCR in various concentrations for24,48and72h,respectively.The increasement of inhibited rates of the proliferation increased with increasement of concentration and incubative time.The IC 50 values at24,48,72h were(2.01±0.26)μg/ml,(1.59±0.23)μg/ml and(0.92±0.11)μg/ml respectively.The difference among IC 50 values was significant(P0.01).When the cells were treated with VCR on0.5,1and2μg/ml for6,12,24h,respectively,the number of the cells in G 0 /G 1 phase decreased remarkably but increased obviously in G 2 /M phase(P0.01).The longer incubative time was,the more variation.[Conclusion]VCR can inhibit the proliferation of CNE-2Z cells.The inhibition depends on dose and incubative.VCR can arrest CNE-2Z cells in G 2 /M phase,it is also in dependence on incubative time.
