Abstract
[Objective] The study aimed to discuss the optimum technology condition for the ultrasonic microwave assisted extraction of polysaccharides from Inonotus obliquus.[Method] With water as the extracting solvent, the polysaccharides from I.obliquus were extracted by ultrasonic microwave assisted extraction technology. The effects of microwave power, microwave treating time and material-liquid ratio on the yield and purity of polysaccharides from I.obliquus were investigated through the response surface analysis. The technology parameters for extracting polysaccharides from I.obliquus by ultrasonic microwave assisted extraction were optimized and they were compared with that by traditional hot water extraction.[Result] The optimum technology condition for ultrasonic microwave assisted extraction of polysaccharides from I.obliquus was determined as follows:the extraction time ranged from 18.45 min to 24.50 min,the material-liquid was 1∶20 and the microwave power ranged from 88.3 W to 96.7 W.Compared with the traditional hot water extraction, the ultrasonic microwave assisted extraction could shorten the extraction time greatly and increase the yield from 2.12% to 3.25% as well as the purity from 64.03% to 73.16%.[Conclusion] Compared with the traditional hot water extraction, the ultrasonic microwave assisted extraction could not only shorten the extraction time greatly, but also increase the yield and purity of polysaccharides from I.obliquus.
