Abstract
Objective:To study the activity of NF-kB in human umbilical vein endothelial cell(HUVEC),HUVEC induced by TNF-α and the relationship between NF-kB and mitogen-activated protein kinase(MAPK)pathway.Methods:After the cultured HUVEC was treated with TNF-α,the activity of NF-kB in HUVEC was measured by electrophoretic mobility shift assay(EMSA)and the expression of IκBα was measured by western blotting.In addition,PD098059(an inhibitor of ERK),SP600125(an inhibitor of JNK),SB203850(an inhibitor of p38MAPK)and PDTC(an inhibitor of NF-kB)were evaluated for their influence on the activity of NF-kB in cultured HUVEC induced by TNF-α.Results:In HUEVC induced by 10ng/ml of TNF-α,the activity of NF-kB was increased in time-dependent manner,at the same time,the expression of IκBα were decreased in time-dependent manner;PD098059 and PDTC,but not SP600125 and SB203850,attenuated the activity of NF-kB in HUEVC induced by TNF-α.Conclusion:In HUEVC,TNF-α can induce NF-kB activation through degraded IκBα;moreover,p38MAPK,but not ERK and JNK,is involved in TNF-induced NF-kB trans-activation.
