Abstract
Objective:To investigate the possible pathogenesis of gastric mucosa injury in rats with duodenogastric reflux(DGR).Methods:Sixteen adult SD rats were undergone surgery.The common hepatic duct and duodenum of SD rats were intubated to collect bile(n=8)and duodenal mixed juice(including bile,pancreatic and duodenal juice,n=8)respectively.Twenty-four SD rats were randomly divided into group A,B and C.Group A and B were different DGR model group.Group A was transfused with bile to stomach,group B was with duodenal mixed juice and group C was with 0.9% saline.The rats were executed after 14 days.Observing pathology of gastric mucosa by HE staining.Immunohistochemical staining was used to detect the expression of cytokines in gastric mucosa.Cell apoptosis was determined by TUNEL technique.Results:The pathology of gastric mucosa in both group A and B demonstrated that rat models with DGR were successful.The expression index of TNF-α and IL-1β in group both A and B were significantly higher than those in control group(P0.05),while there was no significant difference in IL-4 among the three groups(P0.05).The apoptosis cells could be seen intensively in mucosa in DGR model group.Apoptosis index in group both A and B were significantly higher than that of group C(P0.05).No significant difference was found in the pathology,expression of cytokine and cell apoptosis between group A and B(P0.05).Conclusion:Different components in DGR induce gastric mucosa injury,over expression of TNF-α and IL-1β as well as cell apoptosis may play important roles in the pathogenesis of gastric mucosal injury with DGR.
