Abstract
Object To prepare the organophosphate chlorpyrifos artificial antigen,so as to provide technical support for the development of ELISA.Methods The hapten of the organophosphorus insecticide chlorpyrifos was synthesized by using chlorpyrifos reacted with 3-marcapropanoic acid in mild alkaline solution.The hapten was conjugated to bovine serum albumin(BSA),and ovalbumin(OVA)respectively by EDC method.The former resulted in artificial immunoantigen,and the latter artificial coating antigen.Results The molecular weight of synthesized hapten by ESI was 419.7,the same as the theoretical value(420).The ratio of hapten conjugated to BSA,and OVA was 13∶1,and 5∶1 respectively.Conclusion The hapten and artificial antigen were obtained which provides the basis for developing an enzyme-linked immunosorbent assay for chlorpyrifos.
