Abstract
Objective To observe the effect of anti-fibrosis drug on the concentration of cefotaxim in bronchioalveolar lavage fluid.Methods 120 Wistar male rats were randomly divided into five groups,including normal control group(Group A),model group(Group B),interferon-γ-treated group(Group C),ambroxol-treated group(Group D)and Dexamethason-treated group(Group E).On experimental day(day 0),the rats were intratracheally instilled with bleomycin(5 mg/kg)or sterile saline,and then treated with the interferon-γ,ambroxol(35 mg/kg),Dexamethason(0.5 mg/kg)or sterile saline intraperitoneally respectively.On day 7,14,28 after instillation,six rats of each group were sacrificed and the bronchialalveolar fluid was recovered and the lungs were got for histopathological examination after injected cefotaxim(600 mg/kg or 1 200 mg/kg)intravenously.The concentration of cefotaxim in the bronchialalveolar fluid was assayed by liquid chromatography-mass spectrometry method.Results On day 7,the concentrations of cefotaxim in the bronchialalveolar fluid of Group D,Group E were lower than Group B and those of Group C higher than Group B.The difference between Group E and Group B was significant statistically(P0.05).On day 14,the concentration of cefotaxim in the bronchialalveolar fluid of Group D,Group E increased,while Group C decreased.The three groups were all higher than Group B.The difference between Group D and Group B was significant statistically(P0.01).On day 28,the concentration of cefotaxim in the bronchialalveolar fluid of Group E increased continuously,which was significantly higher than other groups.Group C and Group D decreased continuously,and the concentration of cefotaxim of both groups was lower than Group B.Conclusion Dexamethason can increase the concentration of cefotaxim in the bronchialalveolar fluid of the fibrosis stage,while interferon-γ and ambroxol can not.
