Isolation and sequence analysis of a chalcone synthase gene OvCHS from Orychophragmus violaceus

Abstract

PCR primers were designed according to known gene sequences registered in GenBank.A chalcone synthase gene,designated OvCHS,was isolated from petal genomic DNA and cDNA of Orychophragmus violaceus,respectively.Gene sequence was submitted to NCBI GenBank with accession number of EF408918.Sequence analysis showed that genomic DNA of OvCHS was 1263 bp with an intron of 75 bp and a complete coding se-quence of 1188 bp in length encoding 395 amino acids.Sequence comparison with model organism Arabidopsis thaliana indicated that there were 135 bp and 16 amino acids residues variance in coding sequences and protein sequences with similarities of 88.64% and 95.95%,respectively.
