Optimization of Subculture Conditions of Eucommia ulmoides Callus

Abstract

To establish regeneration system of Eucommia ulmoides callus,the buff callus from mature embryos and cream white callus from young embryos were used to study the effects of medium,subculture time,and subculture times on the growth of callus and induction of adventitious bud.The results showed that buff callus had the ability of differentiation after subculture,the cream white callus from young embryos died after subculture.The suitable subculture medium for callus was MS+6-BA 1.5 mg/L+NAA 0.05mg/L and suitable subculture period was 18days.The multiplication of callus was significantly affected by the subculture times,the multiplication times was highest with 2.5after the subculture for four generations,the induction of adventitious bud was also significantly affected by subculture times,the induction rate of adventitious bud was highest with 66.7%after the subculture for two generations,and the multiplication coefficient was highest with 3.2after the subculture for four generations.
