Abstract

Objective To investigate the changes of interleukin-6(IL-6) and tumor necrosis factor-α mRNA in frontal lobe and IL-6 and TNF-α levels in serum of rat diffused truamatic brain injury model.Methods Mamarou model of rat diffused traumatic brain injury was made,venous blood and frontal lobe were collected after different time course of injury.RT-PCR and radioimmunoassay was used.Results IL-6 mRNA level in frontal lobe increased and reached peak 6 hours after diffused traumatic brain inury.IL-6 level in serum also elevated and reached peak 12 hours after diffused traumatic injury compared to control group.TNF-α mRNA expression in frontal lobe was up-regulated and reached peak 12 hours after injury,TNF-α level in serum elevated gradually until 24 hours after injury compared to control.Conclusion Induction of upregulation of IL-6 and TNF-αmRNA expression in frontal lobe and elevation of their protein levels in serum by diffused traumatic brain injury indicate that IL-6 and TNF-α may play important role in the process of diffused traumatic brain injury.
