Optimal conditions of tannase production from Aspergillus niger B0201 undersubmerged fermentation

Abstract

Objective: The conditions of tannase production from Aspergillus niger B0201 under submerged fermentation was studied.Methods: The activity of tannase was determined by ultraviolet spectrophotometric method.The production conditions of tannase from A.niger B0201 were optimized by single factor method.Results: Intracellular enzymes were mainly produced during submerged fermentation.The optimal medium for enzyme production contained 4% tannins,2.5% glucose and 1.25%(NH4)2SO4.The optimal conditions for tannase production were as follows: temperature 30℃,initial pH 5.0,inoculum 1×108/ml spores,medium volume 50ml,rotating speed 140 r/min.Under above conditions,the activity of tannase was highest and reached 145.2 U/ml after 72 h fermentation.Conclusion: Under optimal conditions,the activity of intracelllar tannase was relatively high.The fermentation processing was simple and easy to control.The results might provide important foundation and better guidance for industrial production of tannase.
