Abstract

Objective:In order to elucidate the mechanism of TMP inhibiting VSMC proliferation,we examined the AP-1 and PCNA protein expression in VSMC induced by PDGF.Methods:Cultured cells proliferating model of rats' VSMCs were established by PDGF.Cultured cells were divided into three groups: control group、PDGF group and TMP(low dose,middle dose,high dose)group.The number of VSMC were accounted under microscope.The expression of activator protein-1(AP-1) and proliferating cell nuclear antigen(PCNA)were analysed by Western blotting respectively.Results:The proliferation of rat VSMCs induced by PDGF can be inhibited by TMP significantly in a dose-dependent manner.A remarkable decrease of AP-1 and PCNA expression were demonstrated in the TMP group compared with the model group.Conclusion:TMP inhibits VSMC proliferation associated with down regulation of AP-1 and PCNA expression.
