Abstract

Objective To investigate the effects of platelet-rich plasma (PRP) on the proliferative and capillary canaliculization of human umbilical vascular endothelial cells in vitro. Methods PRP was prepared by twice centrifugal method, and HUVECs (CEV-304) were cultured in vitro and then randomly divided into experimental group and control group. Special culturing media including 5%, 10%, and 20% PRP were added into the HUVECs separately in the experimental group and none in the control group. The proliferation of HUVECs was measured by XTT assay, the cell cycle was tested by flow cytometry, and the ability of capillary canaliculization of HUVECs was tested by tube formation assay. Results HUVECs in experimental group had better shape and higher density than those in control group. XTT method showed that the proliferative activity of HUVECs in experimental group was obviously stronger than that in control group (P 0.01). As compared to the cells in control group, those in S-phase were increased in experimental group (P 0.01), and formed a complete canaliculization and the area of canaliculus was significantly bigger (P 0.05). Conculusions PRP can promote the proliferation and the capillary canaliculization of HUVECs in vitro, and may promote the angiogenesis in the process of tissue regeneration.
