Abstract

Objective: To investigate the effect of glucose on the proliferation of newborn rat mandibular osteoblasts(NRMOBs).Methods: The NRMOBs were cultured in vitro.The effect of different doses of glucose concentration(5.5、 16.5 and 49.5mmol/L) on NRMOBs was investigated.The NRMOBs proliferation was examined after 1、 3、 5、 7、 9 days of culture using Methylthiazol Tetrazolium(MTT).Results: The NRMOBs positively expressed vimentin and osteocalcin,negatively expressed keratin,secreted alkaline phosphatase(ALP),and produced calcium nodules.At 1th day of culture,16.5mM glucose decreased slightly NRMOBs proliferation,and 49.5mM glucose significantly decreased NRMOBs proliferation.The 16.5mM and 49.5mM glucose started to increase NRMOBs proliferation at 3th day of culture,compared to 5.5mM glucose group,the differences had no significance.At 5th、7th、9th day of culture,16.5mM glucose significantly increased NRMOBs proliferation.The 49.5mM glucose started significantly to increase NRMOBs proliferation at 9th day of culture.Conclusion: This study suggests that glucose concentration affects NRMOBs proliferation.Early stage,high-glucose inhibits the proliferation of NRMOBs.Later period,high-glucose gradually promotes the proliferation of NRMOBs.
