Abstract
Objective To study the specific silencing effect of siRNA targeting MYCT-1 gene on cell apoptosis, proliferation and cell cycle of GES-1 cells.Methods The chemically synthetic siRNA targeting MYCT-1 was transfected into GES-1 cells, then flow cytometry were used to detect apoptosis and cell cycle of target cells.Methyltetrazolium(MTT) assay was applied in measuring cell proliferation.Results GES-1 cells transfected with the MYCT-1-RNAi were found to exhibit significantly higher apoptosis and less proliferation than empty vector transfectants.The target cells stagnated in phase S after transfection.Conclusion It was necessary to maintain certain expression level of MYCT-1 gene for GES-1 cells implementing growth control.The regulation of MYCT-1 gene to cell cycle occurred in the conversion process phase S to phase G1.
