Antiproliferation effect of total alkaloid fraction of Yanhusuo on six human gastric cancer cell lines in vitro

Abstract

[Objective] To investigate the antiproliferation effect of total alkaloid fraction of Yanhusuo on six human gastric cancer cell lines as well as its impact on apoptosis and cell cycle phase of MKN-28 cells after treatment;[Methods] MTT assay was used to assess the proliferation of the six cell lines treated with the total alkaloid.The apoptosis and cell cycle phase of MKN28 cells was determined by Flow Cytometer(FCM).[Results] The total alkaloid significantly inhibited the proliferation of the six human gastric cancer cell lines,especially AGS and MKN 28,in a concentration-dependent manner in vitro.The result of FCM in the range of 25 μg/ml and 100 μg/ml for 24h on MKN-28 cells showed higher apoptosis rates(17.2% and 32.3%,respectively),with a significant difference compared with that of the control group(P 0.05).The ratio of S phase was significantly increased at 25 μg/ml,but there was no difference in cell cycle phase after exposure to 100 μg/ml compared with the control group.[Conclusion] Our results suggested that total alkaloid fraction of Yanhusuo has significant antiproliferation effect on six human gastric cancer cell lines and the mechanism is probably related with its regulatory effects on cell cycle and apoptosis in cancer cells.
