Succinic acid production by facultative anaerobic fermentation from corn fiber

Abstract

Using enzymatic-acid two-steps hydrolysis of corn fiber,inspected in dilute HCl hydrolysis conditions on the yield of reducing sugar.And it focused on the study of the growth and succinic acid production by Actinobacillus Succinogenes in facultative anaerobic fermentation.It discussed the feasibility of replacement of glucouse by corn fiber as the carbon source for fermentation succinic acid.The optimal values of the variables for hydrolysis were found to be:1.5% HCl,4 h,100 ℃ and 12% substrate,respectively,and the overall sugar yield in the hydrolysate exceeded 83%.A satisfactory result for fermentation could be obtained with the concentration of reduce sugars was 40 g/L,using corn steep liquor as the nitrogen source,fermentation temperature was 35 ℃ and fermentation time was 60 h on cell growth and succinate production.Under these conditions,succinic acid yield reached to 19.41 g/L.Fermentation of succinic acid using corn fiber and corn steep liquor which is by-products of corn processing as raw material has a good application prospect.
