The Research on the Protection of Anti-ICAM-1 Antibody to the Ischemical Reperfusion Injury of Rats by Nasal

Abstract

Objective To approach the protection of administration of anti-ICAM-1 antibody via nasal to rat cerebral ischemia perfusion.Methods Thirty male Wistar rats were divided into 5 groups: the sham operation group; cerebral ischemia reperfusion(CIR) group;CIR and high,middle and low dose antibody group.The CIR model was made by inserting a suture into the middle cerebral artery(MCA).The high,middle and low dose anti-ICAM-1 IgGs were given to the later groups via nasal delivery after having been ischemia 1 hour.After reperfused 24 hour the rat brains were carried out into sections in coronal colored by TTC,HE and immunohistochemistry of ICAM-1.The volume of cerebral infarction and the ratio of immunohistochemistry positive area of ICAM-1 in all were recored.Results Meninges hyperemia and brain tissue swelling were reveled in the ischemia side of ICA,ICA and low,middle,and hight anti-ICAM-1 IgGs groups.The cerebral infarction volumes of CIR,CIR and low,middle,hight dose anti-ICAM-1 IgGs groups are 57 042±12 483,32 871±4 325,25 932±3 103 and 15 325±3 356 pixel.The ratios of immunohistochemistry positive area in the all area of these groups were(6.64±0.476)%,(5.15±0.987)%,(4.36±0.682)% and(3.42±0.537)%.The differences among these groups showed a statistical significance.Conclusions (1)The expression of ICAM-1 increases significantly after focal CIR.(2) Nasal delivery of anti-ICAM-1 IgGs may suppress the expression of ICAM-1,and decrease the cerbral infarction volume.(3)The dose of anti-ICAM-1 IgGs via nasal administration is negatively correlated with the infarction volume.(4)The ratio of immunohistochemistry positive area of ICAM-1 is positively correlated with the cerebral infarction volume.
