Effects of TMP on expression of MMP in idiopathic pulmonary fibrosis

Abstract

Objective To investigate the influence of TMP on bleomycin-induced pulmonary fibrosis in rats and explore its mechanism.Methods 80 SD rats were randomly divided into four groups: sham group,model group,prednisone group and TMP group.The rats were received injection with a single dose of bleomycin by trachea except sham group,and rats in sham group with same volume normal saline.One day after the injection,prednisone(1.5mg·kg-1·d-1) and TMP(20mg·kg-1·d-1) were respectively given to those in the prednisone group and TMP group daily by gastrogavage,while equal volume of saline was given to those in sham group and model group.On the 14th,28th day,10 rats per group were sacrificed and the lung was incised to make pathological sections which were stained with HE and Mallory.The expression of MMP-1 in lung was determined with immunohistochemisty.Results TMP could inhibit the airsacculitis and pulmonary fibrosis and reduce the expression of MMP-1 in lung tissue.Conclusion TMP can reduce the extent of alveolitis and fibrosis in bleomycin-induced pulmonary fibrosis and repress the expression of MMP-1,which may be relevant to the mechanism of its protective effects on IPF.
