Abstract

Objective:To evaluate the therapeutic effect s and mechanisms of Huayulifei on blemycin-induced pulmonary interstitial fibrosis in rats.Methods:The rats were randomly divided into control group,model group,Huayulifei group and hydrocortisone group. Rat pulmonary fibrosis was induced by blemycin(5mg/kg) in experimental groups,while 0.9% saline in control group.Treatments were started from the next day with a daily oral Huayulifei(13410mg/kg)in Huayulifei group,intraperitoneal Hydrocortisone sodium succinate(25mg/kg)in Hydrocortisone group,and oral saline in control group and model group,respectively.Rats were killed at14,and 28 days after bleomycin, lungs isolated, Hematoxylin and eosin-staining,and expression of Smad3、Smad7 protein was measured by immunohistochemical technique, the activities of MMP2/9 was measured by gelatin zymography.Results: In the Huayulifei group having little inflammatory cells,and the alveoli structure was nearly normal,and the most of the airsacculitis and fibrosis of lung was mild. In Huayulifei group the expression of Smad3 protein was significantly lighter than that in model group and hydrocortisone group,while the expression of Smad7 protein was higher;the activities of MMP2/9 was significantly ligher than that in model group and hydrocortisone group.Conclusion: Huayulifei can ameliorate blemycin-induced pulmonary fibrosis in rats,and It may partly work by affecting the expression of Smad signal pathway in lung tissue to decrease the activation of MMP2/9.
