Establishment of Liver Fibrosis Model Induced by Thioacetamide and its Diagnosis Discriminant Models of the Serum Indexes

Abstract

To produce a reliable and reproducible animal model on hepatic fibrosis which was induced by individual dose of thioacetamide(TAA),and to observe the pathological characteristics of liver fibrosis in Sprague-Dawley (SD) rats,the relationship of liver fibrosis classification and the serum indexes was analyzed preliminarily and the mathematical models were built for diagnosing liver fibrosis.Male SD rats were divided into normal, control and model subgroup respectively.All rats were observed at 8th week and 12th week after TAA intoxication. Rats in the model subgroup were injected intraperitoneally(i.p) with TAA 3 consecutive days per week and lasting for 8 weeks at a dose of 6%TAA 200mg/kg as initial dose.Typical liver fibrosis was induced after 8 weeks of TAA,with the hepatofibrosis pathological characteristics of having fibroblast extending around from central venous or portal area,forming the obvious fiber separator without the formation of false lobules.The dead rats were about 16.0%,and the rate of liver fibrosis model establishment was 90.0%.There was reciprocal change trend to LN,Collagen TypeⅠ,Alb and A/T.The correctly classified total rate of the original grouped cases was 90.5%.The susceptibility and specificity predictive value by the discriminant models for diagnosing liver fibrosis were 84.6%and 100%respectively.The discriminant models are sensitive for predicting the liver fibrosis rat model induced by TAA.
