Effect of Two Different lymphocyte Separatory Liquids on the Extraction of Umbilical Cord Blood mononuclear cells and Later Induction Culture of CIK Cells

Abstract

Objective:To compare the result of umbilical cord blood mononuclear cells extracted by two different lymphocyte separation liquids,and the later effect on CIK culture.Methods:The improved methods were used to extract mononuclear cells from umbilical cord blood.Then the mononuclear cells were inducted and cultured to CIK cells.Lastly,the blood corpuscle count plate and trypan blue were used to determination the density and live rate of CIK cells.Results:Compared with the GE healthcare,the quantity of mononuclear cells separated by TBD was less,while the stratification was clearer and more convenient to extract.Besides,when the cells cultured to CIK later,their density and live rate were close gradually.Conclusion:The TBD lymphocyte separation fluid was more economical and convenient than GE healthcare when used to separate mononuclear cells from umbilical cord blood;in addition,there were no significant difference on later culture.
