Isolation and Identification of Aeromonas veronii from Fish

Abstract

To construct the molecular identification by 16SrRNA gene of Aeromonas veronii from the market fish,the commercial crucian carps were used as the experimental animals,two bacterial strains were isolated from the gills and intestinal tract of crucian carp(Carassius carassius),and were named CC1201,CC1202,respectively.Then two strains were identified by using 16Sr RNA gene sequence and phylogenetic tree analysis of CC1201 from gills and CC1202 from intestinal tract.Furthermore,PCR technology was applied to amplify 16Sr RNA gene sequences of the two isolated strains,and obtained the fragment sizes are 1509bp,of which five bases are different between CC1201and CC1202;the phylogenetic tree analysis showed that CC1201 was most closely related to Aeromonas veronii(FJ940848),the sequence homology was 99.87%,CC1202 was most closely related to Aeromonas veronii(FJ940827),the sequence homology was 99.93%.The strains CC1201 and CC1202 were both identified as Aeromonas veronii.The result may provide a scientific reference for the quarantine inspection of aquatic Aeromonas veronii as well as the diagnosis of diseases in fish.
