Construction of Transgenic Mice Carrying Enhanced Green Fluorescent Protein Gene by microinjection

Abstract
It was attempted to develop green fluorescent protein(GFP) transgenic mouse and produce its lineages. GFP gene was introduced into the zygote of FVB/NJ mice by microinjection. The tail of GFP mice obtained from the founders of three mouths mice and picked-up the genome of mice, genomic DNA was extracted from the offspring of the founder mice for PCR and Southern blotting analysis. GFP gene was injected into pronucleus of 1 586 zygotes and 386 injected eggs were implanted into 32 foster mothers, 4 of which were pregnant and bore 13 off springs. Among the babies, 4 offsprings were integrated with the GFP gene and expressed it. The embryo of 2 founders mice were observed for fluorescence microscopy observation and succeed to detect GFP expression in the embryo of the founder mice. The model of transgenic GFP was established primarily.
