Tacrolimus pretreatment on renal ischemia-reperfusion injury Bcl_2 protein expression

Abstract

Objective Tacrolimus pretreatment on renal ischemia-reperfusion injury Bcl_2 protein expression were discussed.Methods Election 60 healthy Wistar rats were assigned randomly into three groups:sham operation,control group,experimental group.In each group there were 20 rats.Sham operation,only open abdominal,separation of renal artery,no blocking blood flow;control group,Ischemia in the past 6h from the end of intravenous injection of saline 1 ml;experimental group,Ischemia in the past 6h from the end of intravenous injection of Tacrolimus(0.5mg/kg).Reperfusion at different points in time when the blood and kidney samples were removed from rats,serum creatinine(scr),blood urea nitrogen(BUN) weredetermined by automatic biochemical analyzer;TNF-a serum levels were determined by ELISA;Detection of renal Bcl-2 was detected by immunohistochemistry of(sp);kidney injury was Analysised HE staining.Results In reperfusion 2h,6h and 24h,the experimental group of serum creatinine(scr),blood urea nitrogen(BUN) level was significantly lower than the control group(p 0.05),Instead,Bcl-2 immune response intensity significantly higher than the control group(p 0.05).By the light microscope,Control group of renal tubular epithelial cells and the degree of emphasis on disease;The experimental group of renal disease and renal epithelial cells than the control group reduced significantly.Sham group of tubular and tubular epithelial cells no obvious change.Conclusion Tacrolimus on renal ischemia-reperfusion protective effect that may be through increased renal expression of bcl-2 protein,lead to improvements in renal ischemia-reperfusion injury.
