Abstract
Objective To study the influences of Ganxinning Decoction on protein expressions of α-smooth muscle actin(α-SMA),matrix metalloproteinase-13(MMP-13) and tissue inhibitor of metalloproteinase-1(TIMP-1) of liver tissue in rats with hepatic fibrosis.Method All male SD rats(n=68) were randomly divided into the normal group,model group,colchicines group,high-dose Ganxinning Decoction group and low-dose Ganxinning Decoction group.The rat model of hepatic fibrosis was established by the hypodermic injection of carbon tetrachloride(CCl4) and feeding by high-fat low-protein feed and 30% alcohol.At the end of the 6th week the expressions of α-SMA,MMP-13 and TIMP-1 were detected by using immunohistochemical stain method.Result The expressions of α -SMA and TIMP -1 decreased significantly in high -dose Ganxinning Decoction group,low -dose Ganxinning Decoction group and colchicines group compared with those in the model group(P0.05).The expressions of α-SMA and TIMP-1 decreased significantly in high-dose Ganxinning Decoction group and low-dose Ganxinning Decoction group compared with those in colchicines group(P0.05).The expression of MMP-13 in high-dose Ganxinning Decoction group,low-dose Ganxinning Decoction group and colchicines group increased significantly compared with that in the model group(P0.05).The expression of MMP-13 in high -dose Ganxinning Decoction group and low -dose Ganxinning Decoction group increased significantly compared with that in colchicines group(P0.05).Conclusion The inhibitory effect of Ganxinning Decoction on hepatic fibrosis may be related to that it can reduce the protein expressions of α-SMA and TIMP-1,and promote MMP-13 expression.
