Abstract
Objective: To study the effects of Gexiazhuyu decoction on established rat liver fibrosis,and examine its effects on the expression of matrix metalloproteinase-9(MMP-9) and tissue inhibitors of metalloproteinase-2(TIMP-2).Methods: Sixty male Sprague-Dawley rats were randomly divided into four groups as follows: blank control group,Gexiazhuyu decoction group,porcine serum group,porcine serum plus Gexiazhuyu decoction group.All rats were sacrificed at the end of experiment,and liver were collected for analysis.The hydroxyproline(Hyp) content in liver tissue was detected by spectrophotography.The content of MMP-9 and TIMP-2 in the liver was detected by enzyme linked immunosorbent assay.MMP-9 and TIMP-2 mRNA levels in the liver was detected by real-time reverse transcriptase polymerase chain reaction.Results: Compared with the porcine serum group,Hyp content in liver tissue of rats given Gexiazhuyu decoction decreased significantly.mRNA expressions and protein content of MMP-9 were elevated significantly,and TIMP-2 content in liver tissue was decreased significantly in porcine serum plus Gexiazhuyu decoction group.Conclusion: Gexiazhuyu decoction may exert anti-hepatic fibrosis effect in rats via regulation of MMP-9 and TIMP-2 gene expression.
