Abstract
Objective To observe the roles of rosiglitazone in unilateral ureteral obstruction rats renal interstitial fibrosis.Methods Rats were randomly divided into rosiglitazone-treated group,UUO model group and sham-operated group,respectively.The rats in the rosiglitazone-treated group and UUO model group underwent UUO.After 14 days of the operation,all rats were sacrificed and the resected kidneys were collected for HE and MASSON staining and immunohisto chemical staining for CTGF and α-SMA.Results In the obstructed kidneys,there was significant renal interstieial fibrosis and interstitial space expansion.Immunohistochemical staining showed that the expression of CTGF and α-SMA was significantly increased in the obstructed kidneys.In comparison with the model group,rosiglitazone could obviously decrease interstitial volume and the expression of CTGF and α-SMA.It also improved the histological changes of the UUO renal tissue.Conclusions Rosiglitazone can down-regulate the expression CTGF and α-SMA in UUO rats.This demonstrated its potential ability to inhibit renal interstitial fibrosis caused by UUO in rats.
