Abstract
Objective To study the effects of peroxisome proliferator-activated receptor gamma(PPARγ) agonist rosiglitazone on expression of transforming growth factor β1(TGF-β1) and connective tissue growth factor(CTGF) in the renal interstitum of rats with unilateral ureteral obstruction(UUO). Methods Thirty female Wistar rats were randomly divided into rosiglitazone treated group,UUO model group and sham operated group.The rats of treated group and UUO model group underwent unilateral ureteral obstraction.After the 21st day of the operation,all animals were sacrificed and the operated kidneys were collected for HE and Masson staining and immunohistochemical staining for TGF-β1 and CTGF. Results In obstructed kidneys,there were significant renal interstitial fibrosis and interstitial space expansion.Immunohistochemical staining showed that the expression of TGF-β1 and CTGF significantly increased in the obstructed kidneys. Conclusion PPARγ agonist rosiglitazone can down-regulate expression of TGF-β1 and CTGF in UUO rats.This demonstrates its potential ability to inhibit renal interstitial fibrosis caused by UUO rats.
