Abstract
Objective To study the effect of rosiglitazone(RGZ) on the expression of monocyte chemoattractant protein-1(MCP-1)and vascular cell adhesion molecule-1(VCAM-1) mRNA and protein in human umbilical vein endothelial cells(HUVECs)cultured under high-glucose and C-reactive protein(CRP) environment.Methods HUVECs were cultured in vitro and MTT examination showed that RGZ at 5.0 μmol/L had little effect on HUVEC activity.HUVECs at the 5th generation were divided into four groups: control group(C),high glucose group(HG),HG plus CRP group(HGC),CRP group(CRP).The cells were treated with RGZ(5.0 μmol/L) for 24 hours except the control group.mRNA expression of MCP-1 and VCAM-1 was determined by real-time PCR;the proteins of MCP-1 and VCAM-1 were determined by Western blot.Results The mRNA and protein expression levels of MCP-1 and VCAM-1 in HUVECs of HG,HGC and CRP groups were higher than that of C group(P0.01).The mRNA and protein expression levels of MCP-1 and VCAM-1 in cells treated with RGZ were lower than before treatment(P0.01).Conclusion RGZ plays a key role in delaying atherosclerosis by decreasing the expression of MCP-1 and VCAM-1.
