Abstract
Objective To investigate the effects of LPS on HMGB1 and HBD-1 expression in SW480 and A549cells.Methods Upon the concentration of 5,10,15,100 μg/mL LPS exposure in 24h,the RNA of SW480 and A549 cells were extracted and the HMGB1 and HBD-1 expression level were identified by reverse transcriptase-polymerase chain reaction(RT-PCR).Results Both in SW480 and A549 cells,HMGB1 and HBD-1 had a higher expression.Under the different concentrations of LPS stimulation,in SW480 cells,5 ng/μL LPS could increase the expression of HMGB1 gene mRNA,but its expression level was not increased following the increasing concentration of LPS.Different concentrations seemed not to increase the HBD-1 gene expression levels(P0.05).In A549 cells,various LPS concentrations did not change the expression level of HBD-1 and HMGB1.Conclusion HMGB1 and HBD-1 inherently express in epithelial cells of the gastro-intestinal and respiratory tract.LPS could increase the expression of HMGB1 gene mRNA,but its expression level is not increased following the increasing concentration of LPS,while different concentrations of LPS stimulation have no significant effect on the expression of HBD-1 in the SW480 cells.In A549 cells,different concentrations of LPS stimulation have no significant effects on the expression of HBD-1 and HMGB1.
