Abstract
Objective To investigate the effects of dexmedetomidine(DEX)on human umbilical vein endothelial cells(HUVECS)injury induced by hydrogen peroxide(H2O2).Methods HUVECS were cultured in vitro and randomly divided into 6 groups:NC group,H group,D group,DH group,DHY group and HY group.HUVECs oxidative stress injury models were constructed by 200 μmol /L of H2O2.The differences in cell morphology of HUVECS were observed under inverted microscope and the viablity of HUVECS was determined by the methyl thiazolyl tetrazolium(MTT)assay.Hochest staining and flow cytometry were used to detect HUVECS apoptosis.The activity of superoxide dismutase(SOD)and contents of malonadehyde(MDA)in the supernatant of HUVECS were determined by spectra photometric assay.Results As compared with those in NC group and DH group,the viablity of HUVECS and the activity of SOD were decreased(P 0.01 and P 0.05;both P 0.05,respectively),the apoptosis rate and the content of MDA were increased(both P 0.05;both P 0.05,respectively).Conclusion Dexmedetomidine can inhibit the injury of HUVECS induced by H2O2 and has a protective effect.
