Preparation of Acellular Nerve Allografts and Analysis of Their Components

Abstract

Objective To study the preparation of the acellular nerve allografts and analyze the components. Methods The acellular nerve allografts were prepared with the hypotonic acellular method and the characters of the structure were observed under light and electron microscope, and the components were analyzed by immunohistochemistry and polyacrylamidedel electrophoresis (PAGE).Results Schwann cells, the cells of epineurium or nerve fascicles, myelin sheath and axon of nerve fibers were eliminated, but the basement membrane tube of Schwann cells and three-dimensional supports of extracellular matrix of epineurium and nerve fascicles were reserved for normal peripheral nerves treated acellularly. The component analytical results of the acellular nerve allografts indicate that they contain laminin, fbronectin, growth associated protein GAO-43 and 65kDa protein those can facilitate and induce the regeneration of the injured nerves.Conclusion The acellular nerve allografts contain several, proteins those induce and facilitate the regeneration of the injured nerves.
